Product Description


The Gastric Inhibitory Polypeptide Receptor (GIPR) is a class B G protein-coupled receptor (GPCR) that mediates the physiological actions of the incretin hormone GIP, primarily involved in glucose homeostasis and lipid metabolism. GIPR is predominantly expressed in pancreatic β-cells, adipose tissue, and the central nervous system, where it plays a key role in enhancing insulin secretion in response to nutrient intake.

 

ScreeningBio’s Human GIPR HiBiT cell line is an ideal tool for studying GPCR internalization assay. In this system, N-terminally HiBiT-tagged GPCRs are expressed on the plasma membrane of cells. On addition of exogenous pufified LgBiT, complementation occurs between HiBiT and LgBiT, forming the full-length NanoLuc Lucierase. In the presence of the substrate furimazine, luminescence is read as the experimental output. When treated with an agonist, the HiBiT-GPCR undergoes internalization and is removed from the plasma membrane. Purified LgBiT cannot cross the plasma membrane and complement with the HiBiT-GPCR, and therefore there is less complemented NanoLuc as a result.
