Ion channels are pore-forming proteins that allow the flow of ions across membranes, either plasma membranes, or the membranes of intracellular organelles. Ion channels represent the second largest target for existing drugs after G protein-coupled receptors, the therapeutic areas including arrhythmia, hypertension, local anesthesia, pain, stroke, epilepsy, depression, bipolar disorder, cystic fibrosis, cardiac arrhythmias, COPD, autoimmune disorders, and diabetes. The advent of novel, faster screening techniques for compounds acting on ion channels suggests that these proteins represent promising targets for the development of additional, novel therapeutic agents for the near future.
Ion channels not only constitute a significant class of drug targets, they are also critical for evaluating drug safety including being part of the FDA required safety testing for any drug targeted against human disease.
ScreeNingBio provide functionally validated ion channel products for use in cell-based assays, and an hERG ion channel membrane preps for Fluorescence Polarization (FP) binding studies. These assays are suitable for the compounds profiling and high throughput screening.
Calcium Assays: Fluorescent dye-based assays to measure calcium mobilization in cell lines as a direct indication of Ion channel activation or inhibition.

Nuclear Receptor
Nuclear receptors (NR) are intracellular transcription factors that directly regulate gene expression in response to lipophilic molecules, they affect a wide variety of functions, including fatty acid metabolism, reproductive development, and detoxification of foreign substances.
Humans have 48 nuclear receptors, which when dysregulated are often linked to diseases. Because most nuclear receptors can be selectively activated or inactivated by small molecules, they are prominent therapeutic targets.
ScreeNingBio’s offer the nucleare receptors stable cell lines to support your drug R&D activity
- Reporter assay
- Cell line stablility up to 20 passages

The majority of biological solutes are charged organic or inorganic molecules. Cellular membranes are hydrophobic and, therefore, effective barriers to separate them allowing the formation of gradients, which can be exploited, for example, in the generation of energy. Membrane transporters carry solutes across cell membranes, which would otherwise be impermeable to them. The energy required for active transport processes is obtained from ATP turnover or by exploiting ion gradients.
ScreeNingBio offers stable cell lines to discover the novel compounds targeting transporters.

Signal Pathway
Cell signaling is a process of the cells to perceive and correctly respond to their microenvironment and this ability is the basis of development, tissue repair, and immunity, as well as normal tissue homeostasis. Errors in signaling interactions and cellular information processing may cause diseases such as cancer, autoimmunity, and diabetes.
During drug discovery processes, monitoring the outcomes of targeting cell signaling becomes essential since the readouts can be applied for target identification and validation, as well as lead identification and assessment. Furthermore, the optimization of drug candidates and deep dissection of mechanisms of action require cellular measurements of the signaling pathway activities.
ScreeNingBio offers a comprehensive collection of cell-based pathway indicator assays designed to detect activation or inhibition of complex signal transduction pathways in response to a small molecule or biologic.
Reporter Gene Assays: Luminescence-based assays to monitor the activation or inhibition in signal pathways.
Kinase
Kinases play a central in cell signaling to elicit many physiological responses. Dysregulation of kinase activity leads to many human diseases, especially cancer. A typical signaling pathway involves a kinase cascade to produce a cellular response starting with receptor activation via a ligand-induced activation step. Receptor-specific ligands, such as growth factors, result in receptor tyrosine kinases (RTK) dimerization or oligomerization. This often results in the receptors’ activation and autophosphorylation, allowing them to recruit and activate downstream mediators via their SH2 domains. This event then triggers a signaling cascade that leads to the phosphorylation and activation of downstream kinases to act upon various downstream target molecules, including several cytoplasmic targets and transcriptional factors. The activated targets elicit cellular responses that include the promotion of cell growth and division, protein translation, or other physiological responses.
ScreeNingBio offers an extensive portfolio of functional and binding cell-based assays, cell lines, and recombinant kinase enzymes to evaluate any kinase target-based signaling activities to aid your efforts from discovery through development into commercialization. Screen for novel kinase inhibitors, perform pharmacological profiling, and evaluate kinase activation from therapeutic binding. Utilize cell-based assays to determine therapeutic mechanism of action (MOA) and intracellular compound target engagement. Perform kinase activity assays measuring ADP production or substrate phosphorylation with one of the most extensive offerings of recombinant kinase enzymes available.
- ADP-Glo Assays: Luminescence-based assays to measure ADP production indicating the activation or inhibition of the kinases.
- TR-FRET Assays: TR-FRET based assays to measure the substrate phosphorylation indicating the activation or inhibition of the kinases.

Check Point Cell Lines
Regulation of immune responses is tightly controlled through a balance of co-stimulatory and inhibitory checkpoint receptors often exploited by many cancers. Therefore, therapeutics that block inhibitory receptors or activate immune-stimulatory checkpoint receptors, such as PD-1 receptor, have proven to be powerful agents to restore anti-tumor immune responses.
ScreeNingBio offers a large, diverse selection of assays for characterization, screening, and potency QC lot-release testing of immunotherapy therapeutics targeting both inhibitory and co-stimulatory checkpoint receptors. Utilize functional mechanism of action (MOA)-reflective cell-based checkpoint receptor assays to analyze signaling, dimerization, internalization, and clustering/crosslinking of checkpoint receptors. These simple, rapid assays provide a highly sensitive response for most targets without primary cells or complex protocols in a few hours.
Reporter Cell Lines
Reporter cell lines are stable cell lines that have been labeled with reporter genes, allowing the visualization and tracking of protein expression. The reporter gene is inserted next to the gene promoter of interest within a vector and transfected to a cell line. If the promoter is expressed in the cell line, the reporter gene will also be expressed, allowing monitoring of cellular metabolic processes, transcription factors, and protein localization. Common reporter genes include luciferase and fluorescent proteins like GFP and RFP. The origin of the cell line, intended application, and the reporter should be taken into consideration when selecting your product.
Cas9 Expression Cell Lines
ScreeNingBio’s Ca9 expression stable cell lines constitutively expressing the CRISPR Cas9 nuclease enable you to carry out CRISPR genome editing applications with high efficiency. The Cas9 stable cell lines are available pre-made in many human, mouse, and rat cell line backgrounds. The S.pyogenes CRISPR Cas9 nuclease is stably integrated into either the human AAVS1 or mouse ROSA26 “Safe Harbor” sites, or randomly via lentiviral-mediated transduction.
The CRISPR Cas9 nuclease, in combination with single guide RNAs (sgRNAs), is widely used to create targeted genomic modifications, from gene knockouts, mutagenesis, fusion tagging, and more, in eukaryotic cells and animal models. Stable Cas9 integration ensures robust and consistent expression of the CRISPR Cas9 nuclease.
- Stable Cas9 integration minimizes the need for co-transfection or co-transduction of sgRNAs, ideal for high-throughput sgRNA applications.
- Single clone isolation provides consistent, high-level Cas9 expression in a uniform genetic background.
