Interleukin-6 (IL-6) is a pleiotropic cytokine mainly produced by macrophages, T cells, fibroblasts, and endothelial cells, playing central roles in immune regulation, inflammation, and the acute phase response. IL-6 functions by binding to a receptor complex composed of IL-6Rα (gp80) and gp130, thereby activating downstream signaling pathways including JAK-STAT3, MAPK, and PI3K-Akt. For example, during acute inflammation, IL-6 promotes the production of acute phase proteins such as C-reactive protein (CRP) by the liver and regulates immune cell differentiation and activation, whereas its persistent expression in chronic inflammation is closely associated with autoimmune diseases and tissue damage. Beyond immune modulation, IL-6 is involved in hematopoiesis, bone metabolism, and the shaping of the tumor microenvironment. Dysregulation of IL-6 is frequently observed in various disease states, such as rheumatoid arthritis, Castleman’s disease, cytokine storm, and multiple cancers (e.g., multiple myeloma, breast cancer), where it promotes tumor growth and drug resistance. Consequently, drugs targeting the IL-6 signaling pathway, such as anti-IL-6 receptor monoclonal antibody (tocilizumab) and anti-IL-6 monoclonal antibody (siltuximab), have been successfully developed for the treatment of rheumatoid arthritis, CAR-T‑induced cytokine release syndrome, and other diseases, underscoring the critical role of IL-6 in both normal immune homeostasis and pathogenesis.

 

Screeningbio’s TR-FRET Human IL6 Detection Kit can be used to measure the levels of Human IL6 protein in supernatants. This kit is a sandwich immunoassay developed using TR-FRET technology, characterized by simplicity, rapidity, high specificity, high sensitivity, and good reproducibility.

The basic principle of this method is as follows: a pair of specific antibodies are separately conjugated to TR-FRET Solar Eu*1 (donor) and TR-FRET LA*2 (acceptor). When both antibodies bind to distinct epitopes of the Human IL6 protein, the TR-FRET donor and acceptor are brought into close proximity. Upon excitation by an external light source, fluorescence resonance energy transfer occurs between the donor and acceptor. The level of Human IL6 protein can then be determined by measuring the signal intensity at a specific wavelength (665 nm).
