Product Description


The MCH1 receptor (melanin-concentrating hormone receptor 1) is a G protein-coupled receptor encoded by the MCHR1 gene, belonging to the melanin-concentrating hormone receptor family. It is primarily distributed in the central nervous system, particularly in regions such as the cerebral cortex, hypothalamus, hippocampus, nucleus accumbens, and amygdala, with lower expression levels in peripheral tissues including the eye, skeletal muscle, and tongue. Its core function involves binding the ligand melanin-concentrating hormone and mediating signal transduction via Gi/o and Gq proteins to regulate feeding behavior and energy metabolism, while also participating in the modulation of physiological processes including sleep-wake cycles, emotional responses, learning, and memory. This receptor has emerged as a potential drug target for obesity, depression, and anxiety disorders.

 

Screeningbio’s CHO-K1/Rat MCH1 cell line overexpress rat MCHR1 receptor and is designed to detect inhibition in intracellular cAMP levels or to detect increases in intracellular IP-1 levels in response to agonist stimulation of the receptor. Cisbio HTRF cAMP kit or IP-1 kit can be used to detect the signal.

CHO-K1/Rat MCH1 Agonist Assay. CHO-K1/Rat MCH1 cells were stimulated with 5 μM Forskolin and treated with reference agonist. The assay was run based on Revvity cAMP HTRF protocol. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
