Product Description


The EP4 receptor (Prostaglandin E receptor 4) is one of the four G protein-coupled receptor subtypes for prostaglandin E2, primarily coupling to the Gs protein. It is widely distributed in the cardiovascular system (vascular endothelium, myocardium), kidneys (glomeruli, collecting ducts), gastrointestinal tract, uterus, lungs, bone, and various immune cells. Its core function involves activating adenylyl cyclase to elevate intracellular cAMP levels, thereby mediating vasodilation, inhibition of immune cell activation, promotion of bone formation, gastric mucosal protection, regulation of renal water and sodium excretion, and modulation of inflammation resolution processes. This receptor plays critical roles in conditions such as ductus arteriosus closure, bone metabolic balance, inflammatory bowel disease, and rheumatoid arthritis, making it an important therapeutic target for pulmonary arterial hypertension and inflammatory disorders.

 

Screeningbio’s CHO-K1/Rat EP4 cell line overexpress Rat PTGER4 and is designed to detect increases in intracellular cAMP levels in response to agonist stimulation of the receptor. Cisbio HTRF cAMP kit can be used to detect the signal.

CHO-K1/Rat EP4 Agonist Assay. CHO-K1/Rat EP4 cells were treated with the reference agonist. The assay was run based on Revvity cAMP HTRF protocol. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
