Product Description


Chemokine (C-C motif) receptor 6 (CCR6) is a G protein-coupled receptor whose sole known chemokine ligand is CCL20. It is primarily expressed on Th17 cells, regulatory T cells, B lymphocytes, and specific subsets of dendritic cells. The "CCL20-CCR6" axis serves as a critical pathway for lymphocyte homing to mucosal sites and inflamed tissues, playing a central role in maintaining gut homeostasis, defending against bacterial infections, and driving autoimmune inflammation (such as in psoriasis, rheumatoid arthritis, and inflammatory bowel disease). Consequently, CCR6 is an important therapeutic target for autoimmune and inflammatory diseases.

 

Screeningbio’s CHO-K1/Mouse CCR6 cell line overexpress CCR6 and is designed to detect inhibition in intracellular cAMP levels in response to agonist stimulation of the receptor. Cisbio HTRF cAMP kit can be used to detect the signal.

CHO-K1/Mouse CCR6 Agonist Assay. CHO-K1/Mouse CCR6 cells were stimulated with 5 μM Forskolin and treated with reference agonist. The assay was run based on Revvity cAMP HTRF protocol. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
