Product Description


The RXFP2 receptor (relaxin family peptide receptor 2), also known as LGR8 or GPR106, is a G protein-coupled receptor containing leucine-rich repeats. In humans, it is primarily distributed in the brain, kidney, muscle, testis, thyroid, uterus, as well as peripheral blood cells and bone marrow. Its core function, upon activation by its endogenous ligand relaxin and INSL3 (insulin-like peptide 3), involves stimulating adenylyl cyclase via G proteins to increase intracellular cAMP levels. This receptor plays a critical role in gubernaculum development and testicular descent, and mutations in its gene are a significant cause of cryptorchidism (failure of the testis to descend).

 

Screeningbio’s CHO-K1/Human RXFP2 cell line overexpress RXFP2 and is designed to detect increases in intracellular cAMP levels in response to agonist stimulation of the receptor. Cisbio HTRF cAMP kit can be used to detect the signal.

CHO-K1/Human RXFP2 Agonist Assay. CHO-K1/Human RXFP2 cells were treated with the reference agonist. The assay was run based on Revvity cAMP HTRF protocol. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
