Product Description


The GALCRL-RAMP3 (AM2) receptor is a G protein-coupled receptor complex formed by the calcitonin receptor-like receptor (CALCRL/CLR) and receptor activity-modifying protein 3 (RAMP3). Its distribution in the body differs from that of RAMP2 (i.e., different from the AM1 receptor). Functionally, this receptor primarily recognizes and binds adrenomedullin (AM) and its related ligands with high affinity, playing crucial roles in physiological processes such as regulating vascular tone and fluid balance via the Gs protein-cAMP signaling pathway, as well as in maintaining normal body weight in old age and contributing to immune function.

 

Screeningbio’s CHO-K1/Human GALCRL-RAMP3(AM2) cell line overexpress both CALCRL and RAMP3 accessory protein (also named AM2). The cell line is designed to detect increases in intracellular cAMP levels in response to agonist stimulation of the receptor. Cisbio HTRF cAMP kit can be used to detect the signal.

CHO-K1/Human GALCRL-RAMP3(AM2) Agonist Assay. CHO-K1/Human GALCRL-RAMP3 (AM2) cells were treated with the reference agonist. The assay was run based on Revvity cAMP HTRF protocol. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
