Interleukin-1β (IL-1β) is a potent pro-inflammatory cytokine mainly produced by activated macrophages, monocytes, and dendritic cells. It belongs to the IL-1 family and plays central roles in initiating and amplifying innate immune responses and inflammatory reactions. IL-1β is synthesized as an inactive precursor (pro-IL-1β) and is cleaved by inflammasome-activated caspase-1 to release its mature form. Mature IL-1β functions by binding to the type I IL-1 receptor (IL-1RI) and its co-receptor IL-1RAcP, subsequently recruiting the adaptor protein MyD88 and activating downstream signaling pathways including NF-κB, MAPK, and JNK. For example, during infection or tissue injury, IL-1β rapidly induces fever, promotes endothelial adhesion molecule expression, recruits immune cells, and synergizes with other cytokines to amplify local and systemic inflammation; however, excessive or persistent IL-1β production leads to tissue damage, fibrosis, and septic shock. Beyond inflammation, IL-1β is also involved in hematopoiesis, bone metabolism, and pain sensitization. Dysregulation of IL-1β is frequently observed in various disease states, such as autoinflammatory diseases (e.g., familial Mediterranean fever), rheumatoid arthritis, gout, atherosclerosis, type 2 diabetes, and COVID-19-associated cytokine storm. Consequently, drugs targeting the IL-1 signaling pathway have been successfully developed, including recombinant IL-1 receptor antagonist (anakinra), neutralizing anti-IL-1β monoclonal antibodies (canakinumab, gevokizumab), and an IL-1 trap (rilonacept), for the treatment of rheumatoid arthritis, autoinflammatory diseases, gouty arthritis, and COVID-19-related inflammation, underscoring the critical role of IL-1β in normal host defense, inflammatory homeostasis, and a broad range of inflammatory diseases.
Screeningbio’s IL-1β Detection Kit can be used to measure the levels of Human IL-1β protein in supernatants. This kit is a sandwich immunoassay developed using TR-FRET technology, characterized by simplicity, rapidity, high specificity, high sensitivity, and good reproducibility.
The basic principle of this method is as follows: a pair of specific antibodies are separately conjugated to TR-FRET Solar Eu*1 (donor) and TR-FRET LA*2 (acceptor). When both antibodies bind to distinct epitopes of the Human IL-1β protein, the TR-FRET donor and acceptor are brought into close proximity. Upon excitation by an external light source, fluorescence resonance energy transfer occurs between the donor and acceptor. The level of Human IL-1β protein can then be determined by measuring the signal intensity at a specific wavelength (665 nm).
