Product Description


Interferon-γ (IFNγ) is a pleiotropic cytokine mainly produced by activated T cells and NK cells, playing central roles in cell-mediated immunity, antiviral defense, and antitumor surveillance. IFNγ functions by binding to the IFNGR1/IFNGR2 receptor complex, activating the JAK-STAT1 signaling pathway and inducing the expression of interferon-stimulated genes. Classic functions of IFNγ include activating macrophages, upregulating MHC molecule expression, promoting Th1 differentiation, and enhancing the antitumor activity of CD8⁺ T cells. Dysregulation of IFNγ is associated with various diseases: overproduction leads to autoimmune disorders (e.g., rheumatoid arthritis), whereas defective signaling causes susceptibility to intracellular pathogens (e.g., mycobacteria). Consequently, recombinant IFNγ protein has been used to treat chronic granulomatous disease, underscoring the critical role of IFNγ in both immune defense and pathology.

 

Screeningbio’s IFNγ Detection Kit can be used to measure the levels of Human IFNγ protein in supernatants. This kit is a sandwich immunoassay developed using TR-FRET technology, characterized by simplicity, rapidity, high specificity, high sensitivity, and good reproducibility.

The basic principle of this method is as follows: a pair of specific antibodies are separately conjugated to TR-FRET Solar Eu*1 (donor) and TR-FRET LA*2 (acceptor). When both antibodies bind to distinct epitopes of the Human IFNγ protein, the TR-FRET donor and acceptor are brought into close proximity on excitation by an external light source, fluorescence resonance energy transfer occurs between the donor and acceptor. The level of Human IFNγ protein can then be determined by measuring the signal intensity at a specific wavelength (665 nm).
