Product Description
FYN A, also known as Fyn proto-oncogene isoform A (or simply FYN), is a member of the Src family of non-receptor tyrosine kinases (SFKs) that plays a pivotal role in T-cell receptor (TCR) signaling, neuronal development, cell adhesion, and cytoskeletal remodeling. FYN A is activated upon engagement of cell surface receptors (e.g., TCR, CD28, integrins, and growth factor receptors) through autophosphorylation at Tyr417 (in the activation loop) and interaction with SH2/SH3 domain-binding partners, leading to downstream activation of pathways such as RAS-MAPK, PI3K-AKT, and PLCγ. Beyond its role in normal immune function and central nervous system plasticity, dysregulation of FYN A signaling—through overexpression, aberrant activation, or mutations—is closely associated with T-cell leukemia, Alzheimer's disease (where it mediates tau hyperphosphorylation), melanoma, prostate cancer, and chronic inflammatory diseases, making it a highly validated target for cancer therapy, neurodegenerative disease treatment, and drug discovery.
Screeningbio’s FYN A Kinase Assay Kit provides a robust, sensitive, and high-throughput platform for quantifying FYN A kinase activity and evaluating the potency of tyrosine kinase inhibitors (TKIs).
This kit utilizes the ADP-Glo Kinase Assay platform, a luminescent technology that measures the amount of ADP produced during the kinase reaction.
The assay is performed in two distinct steps:
Kinase Reaction: FYN A catalyzes the transfer of a phosphate group from ATP to a specific substrate, resulting in the production of phosphorylated substrate and ADP.
ADP-Glo™ Reagent Addition: After the kinase reaction, the ADP-Glo™ Reagent is added to terminate the reaction and deplete any remaining unreacted ATP.
Kinase Detection: The Kinase Detection Reagent is added to simultaneously convert the produced ADP back into ATP, which is then used by luciferase to generate a luminescent signal.
The light intensity produced is directly proportional to the ADP concentration and, consequently, to the enzymatic activity of the FYN A kinase.
