Product Description

RARγ (Retinoic Acid Receptor gamma) is the third subtype of the retinoic acid receptor (RAR) family within the nuclear receptor superfamily, alongside RARα and RARβ. It also functions as a ligand-activated transcription factor that binds retinoic acid to regulate gene expression. RARγ exhibits distinct tissue-specific distribution: it is highly expressed in the skin (e.g., epidermal keratinocytes), cartilage, bone tissue, lung, and certain embryonic mesenchymal regions. Its main functions include promoting the differentiation and maturation of epidermal keratinocytes; regulating the development of chondrocytes and osteoblasts, thereby participating in skeletal formation and growth; contributing to limb patterning and the development of skin appendages (e.g., hair follicles) during embryogenesis; and modulating lung epithelial differentiation. Abnormalities in RARγ are associated with certain skin diseases and bone developmental disorders.

Screeningbio’s HEK293/RARγ Luciferase Reporter cell line contains human RARγ gene and a luciferase gene under the control of retinoic acid response elements. The cell line is designed to detect the response to the stimulation of RARγ ligands. Promega One-Glo kit can be used to detect the signal.

Data

HEK293/RARγ Luciferase Reporter Agonist Assay. HEK293/RARγ Luciferase Reporter cells were treated with agonist. The assay was run based on Promega ONE-GLOTM Luciferase Assay System. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
