Product Description


The C3a receptor (complement C3a receptor) is a G protein-coupled receptor that specifically binds the anaphylatoxin C3a. It is primarily expressed on mast cells, basophils, neutrophils, monocytes/macrophages, as well as certain epithelial and smooth muscle cells. Its core function is to mediate the pro-inflammatory effects of C3a by activating Gq/11 and Gi/o proteins to trigger intracellular calcium mobilization, thereby inducing mast cell degranulation, promoting leukocyte chemotaxis, enhancing vascular permeability, and stimulating inflammatory cytokine release. It plays a pivotal role in pathological processes including allergic reactions, sepsis, ischemia-reperfusion injury, and neuroinflammation. This receptor has emerged as a potential therapeutic target for inflammatory diseases and autoimmune disorders.

 

ScreeningBio’s HEK293T/Rat C3a β-arrestin cell line is an ideal tool for studying GPCR/β-arrestin interactions. In this system, the GPCR C-terminus is fused to a smallBiT tag, and the β2-arrestin N-terminus is fused to a largeBiT tag. Upon receptor activation, GPCR/β-arrestin interaction brings the two fragments together to reconstitute an active NanoLuc enzyme, which can be quantified using the NanoBiT substrate. This cell line is designed to evaluate a compound’s ability to activate the β-arrestin signaling pathway.
