The EP3 receptor (Prostaglandin E receptor 3) is one of the four G protein-coupled receptor subtypes for prostaglandin E2, characterized by complex signaling diversity through coupling to multiple G proteins (Gi, Gq, Gs), which can produce different or even opposing biological effects. It is widely distributed in the kidney (collecting ducts, thick ascending limb of Henle), gastrointestinal tract (parietal cells), uterus, vascular smooth muscle, adipose tissue, and the thermoregulatory center in the brain. Its core functions are tissue-specific: in the kidney, it promotes water and sodium reabsorption to regulate blood pressure; in the gastrointestinal tract, it inhibits gastric acid secretion and protects the mucosa; in the uterus and blood vessels, it mediates smooth muscle contraction; in adipose tissue, it suppresses lipolysis; and in the central nervous system, it participates in the negative feedback regulation of febrile responses. This receptor plays a dual regulatory role in hypertension, fever, thrombosis, and inflammatory modulation.
