Product Description


The IP receptor (prostacyclin receptor) is a G protein-coupled receptor activated by PGI2 (prostacyclin), primarily coupling to the Gs protein. It is widely distributed on vascular endothelial and smooth muscle cells, platelets, kidneys (glomeruli), heart, and certain immune cells. Its core function involves elevating intracellular cAMP levels upon activation, thereby mediating potent vasodilation, inhibiting platelet aggregation, protecting the vascular endothelium, regulating renal blood flow, and contributing to anti-inflammatory and anti-proliferative effects. This receptor is an important therapeutic target for pulmonary arterial hypertension (e.g., using iloprost, treprostinil) and thrombosis prevention.

 

Screeningbio’s CHO-K1/Mouse IP cell line overexpress PTGIR and is designed to detect increases in intracellular cAMP levels in response to agonist stimulation of the receptor. Cisbio HTRF cAMP kit can be used to detect the signal.

CHO-K1/Mouse IP Agonist Assay. CHO-K1/Mouse IP cells were treated with the reference agonist. The assay was run based on Revvity cAMP HTRF protocol. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
