Product Description


The SST4 receptor (somatostatin receptor 4) is a member of the five-receptor family for somatostatin and belongs to the Gi/o protein-coupled receptor family. It is widely distributed in the central nervous system, primarily localized on the cell bodies and dendrites of neurons in brain regions such as the cerebral cortex, hippocampus, amygdala, striatum, and hypothalamus. Peripherally, it is also expressed in tissues including the lung (bronchial epithelium, vascular endothelium, and smooth muscle), kidney, and gastrointestinal tract. Its core functions involve inhibiting adenylyl cyclase, modulating MAPK pathways and ion channels, thereby participating in the regulation of neuronal excitability, learning, and memory processes. Additionally, SST4 mediates anti-inflammatory, analgesic, and tissue-protective effects, inhibiting neurogenic inflammation in peripheral organs such as the lungs and potentially contributing to neuroprotection and repair following brain injury.

 

Screeningbio’s CHO-K1/Human SST4 cell line overexpress SSTR4 and is designed to detect inhibition in intracellular cAMP levels in response to agonist stimulation of the receptor. Cisbio HTRF cAMP kit can be used to detect the signal.

CHO-K1/Human SST4 Agonist Assay. CHO-K1/Human SST4 cells were stimulated with 2.5 μM Forskolin and treated with reference agonist. The assay was run based on Revvity cAMP HTRF protocol. Non-linear regression was used to plot activity changes vs. [Compound, M], and EC50 values were determined, using GraphPad Prism software.
